Johann Sebastian Bach was a member of a family that had for generations been occupied in music. His sons were to continue the tradition, providing the foundation of a new style of music that prevailed in the later part of the eighteenth century. Johann Sebastian Bach himself represented the end of an age, the culmination of the Baroque in a magnificent synthesis of Italian melodic invention, French rhythmic dance forms and German contrapuntal mastery.

...It is fascinating to hear the work played on Bruckner’s own instrument and it works surprisingly well. Some of Rogg’s tempi are as quick as I have heard, particularly the adagio, but they make sense in this context.

Moving from the harpsichord to the clavichord or the organ was probably easy enough for Johann Sebastian Bach. The source of the sound didn’t matter, because for the master of Leipzig, what counted were thought and intellect: the form, tonality and melodic contours ere more important than the instrument itself. And indeed, through this work of musical thought, Bach used different keyboards, prefiguring the instruments to come: the piano as a synthesis of the harpsichord, the organ and the clavichord.

Transcriptional Regulation in Eukaryotes: Concepts, Strategies, and Techniques

Michael Carey and Stephen T. Smale, «Transcriptional Regulation in Eukaryotes: Concepts, Strategies, and Techniques»
Cold Spring Harbor Laboratory Press | ISBN 0879696354 | September 2001 | PDF | 671 Pages | 5,1 Mb

The goal of this book is to provide a detailed description of the approaches to be employed and issues to be considered when undertaking a molecular analysis of the transcriptional regulatory mechanisms for a newly isolated gene, or a biochemical analysis of a new transcription factor. Our emphasis is on mammalian transcription, which is complicated by the combinatorial nature of regulation and the lack of facile genetics. We refer periodically to studies in yeast, Drosophila, and other organisms where more tractable genetic approaches have led to a detailed understanding of particular mechanistic issues. The topics covered in the book extend from the determination of whether a gene is in fact regulated at the level of transcription initiation to advanced strategies for characterizing the biochemical mechanism underlying its combinatorial regulation by activators. Although numerous specialized and detailed techniques are included, the unique characteristics of this book are its strategic and conceptual emphasis on analysis of individual genes and the
transcription factors that regulate them. Details
